Yes, unless no one else has played in 48 or 72 hours.  It is also against
the rules to enter a second guess before the first one is scored.

For convenience, we often pause after five unscored guesses.

The latter is not just for convenience but was recommended in the rules in
order to have the clues keep pace with the guesses. This also helps reduce
the possibility of a player winning because they might just be the only
person reading when a larger number of guesses are scored all at once.
