(You've become rather good friends with your supervisor, who happens to be
of the opposite sex. One day, one of your co-workers confronts you. It
becomes readily obvious that this person is insanely jealous of your
relationship, and threatens to tell your spouse that you've been sleeping
with your boss! You know you haven't, but you also know that it's
somewhat plausible. This co-worker won't keep quiet unless you give in to
certain demands.)

I would tell my wife what had happened, and if our relationship wasn't
strong enough for her to believe me then we probably shouldn't have gotten
married in the first place.

what steve said.
if we got married, then we should have a strong relationship.  (this is 
because of me, not any special quality of marriage.  our society seems
to have removed that)  she should be able to trust me enough.  i would
tell her, and trust her to believe me.

I'd tell my spouse, and ask his advice; then I'd tell my boss.
Nobody ever knows *everything* that goes on in somebody else's
mind, and even my spouse-who-trusts-me-completely might feel more
comfortable if the working conditions changed to make the
suggestion less plausible.

each other.

I wouldn't do anything.  My wife wouldn't care if I were having an affair;
she'd probably ask me if it was true or not.  I can see the exchange between
my wife and the work-weasel:  WW "Your husband is having an affair with his 
boss."  Wife "How selfish!  He didn't invite me!"

Well, I would get a ski mask, a baseball bat, and a pair of handcuffs.I would
wait for him one night in the parking lot,wearing the mask, and I would strike
him over the head. When he was sufficiently stunned, I would drag him to a
secluded spot, handcuff him to a lightpost, and while I beat him, I would drop
hints as to the reason,nothing he could take to the police, but enough to know
the reason for all this, let him know that worse would follow should he
persist, and then go home and explain the situation to my wife.(sans beating.)
